Case Study

Enabling Efficient Enzyme Extraction with BPS Disc Centrifuge

Customer Information:

The customer is a leading domestic Porcine Circovirus (PCV) vaccine manufacturer in China. The company specializes in producing PCV vaccines using PK15 cells combined with microcarriers in suspension culture. Their production process emphasizes advanced biotechnological methods and high-quality standards, positioning them as a key player in the veterinary vaccine industry. However, they encountered technical challenges during the harvesting process, and our engineering team provided assistance.


  

Customer Requirements 

The customer required a fully enclosed, sterile, and continuous separation solution to replace their existing inefficient and labor-intensive process. 

Key requirements included:

l A system capable of handling high-volume virus harvesting and separation with continuous feed operation. 

l Superior separation performance: supernatant turbidity ≤200 NTU (improving from 700–1000 NTU) and protein recovery ≥80%.

l Compliance with GMP sterile standards, including full Clean-in-Place (CIP) and Sterilize-in-Place (SIP) capabilities.

l Reduced manual intervention, lower operational costs, and minimized contamination risks.

l Enhanced process efficiency to replace freeze-thaw sedimentation and tubular centrifugation methods.

Customer Challenge 

In March 2025, the customer faced significant challenges during the virus harvesting and separation stage of PCV vaccine production:

l The traditional freeze-thaw sedimentation process required substantial cold storage space and extended processing time, leading to bottlenecks.

l Tubular centrifuges operated in batch mode, resulting in low efficiency, inability to process continuously, and high labor costs.

l The non-closed process posed ongoing contamination risks, compromising product sterility and consistency.

l Existing equipment failed to meet the desired balance between separation efficiency (turbidity reduction) and protein recovery targets

Our Solution

After technical evaluation, we recommended the BPS-L305 disc centrifuge (8500 rpm, separation factor 12,000 G) with full SIP capability and a sterile, enclosed design. This configuration ensures stable, high-efficiency, continuous separation.


  

On-site Test Results

During on-site testing, the equipment demonstrated excellent performance. In the first test, the feed turbidity was 162 NTU, which was reduced to 55 NTU in the supernatant, with a temperature rise of 5°C and protein recovery reaching 99%. In the second test, the feed turbidity was 780 NTU, reduced to 59 NTU in the supernatant, with a temperature rise of 6°C and protein recovery of 80%. It should be noted that the lower protein recovery in the second test was due to different testing conditions and does not reflect a difference in equipment performance.

Visually, the BPS-L305 consistently reduced turbidity well below the target level while maintaining high protein recovery.
